The duo actually takes places in different locations one of which is chosen randomly.

Story

Starro has taken control of heroes and villains alike, and is forcing them to do his bidding. Put a stop to Starro by rescuing those under its influence.

Iceberg Lounge

Deal with the party guests, henchmen and Starro until the first boss, the Pengbot Minimus 1.0, appears on the iceberg. After you defeated him more henchmen and Starro will appear. There will also be mini-icebergs in the pool. Get those to the hatcheries marked in time to avoid Callow Starro to appear. After a while the final boss, Controlled Penguin, will appear. Occasionally you will be covered with Starro Spores. Simply jump into the pool to cleanse yourself of those.
